XL-30 Quick Start These pages give a summary of controls and operation of the microscope. ¥ Inserting a Specimen Settings control area: HT OFF. Wait for beam to go off and for sound of isolation valve closing. VENT. Wait until the chamber is up to atmospheric pressure. This will take 2 minutes - expand Vacuum panel for timer. Do not force door open. Place a specimen into the stage holder and secure. Do not over-tighten. Check, and if necessary adjust, the Z height before closing the chamber door. The tool provided is next to the chamber, and is shown in Figure 5-3 on page 5-7 of the manual. PUMP. Close door and click pump button. Automatic sequence until Vac OK status. Allow vacuum to reach mid -5 range before continuing. Turn on gun voltage. This will open the isolation valve and generate a beam. ¥ Obtaining an Image Menu Bar items: MAGN: 200x or less. BEAM: Spot 3 SCAN: Slow Scan 1 FILTER: Live DETECTOR: SE Settings control area: BEAM: Set KV. HT ON Contrast: Adjust. Brightness: Adjust. Focus with the mouse. ADJ Z to FWD: DO NOT say OK until focus is correct. If needed, increase magnification and refocus on highest point in specimen. Then you can choose Z position (10mm for eucentric position). ¥ Optimizing the Image Menu Bar items: SCAN: Move the specimen into the field of view. MAGN: Select a magnification to suit the visible structure, e.g. 2500x Focus with the mouse. MAGN: Increase the magnification by 2 steps, e.g. 10,000x. BEAM: Select a smaller Spot size, if necessary. Imaging control area: VIDEO: Adjust Contrast and Brightness to suit. FOCUS: Select Window if necessary to see detail. Focus with the mouse. STIGM: Right Button + Shift - correct the astigmatism at Hi-mag. Focus with the mouse. This is an iterative loop: focus - adjust stigmators - focus. Menu Bar items: MAGN: Decrease, 2 steps back, e.g. 2500x. SCAN: Slow Scan 2. View the image ¥ Acquiring a Digital Image from Microscope Control. The image should be focused and stigmated correctly before collecting the image. Contrast and brightness must also be properly adjusted. Menu Bar items: SCAN: Adjust line time & lines/frame for XHD. 9. Reset to defaults at end of session). Set default file name and image number in XL-Docu (SPECIAL Preferences ÓfilenameÓ and # sequence). Set for each image sequence. Switch back to XL Microscope, press F5 to acquire XHD image. This will scan the image once at the setting for Single Scan, then freeze the image. Click the green file cabinet icon to save the image to XL-Docu. ¥ Save images to your folder from within XL DOCU. Transfer images to View Port Window. Select images you want to save in View Port Window. Drag selected images from View Port Window to disk icon below. Confirm file names and location for each file. Save files in folder Users Files 1 week FTP files to REMF file server. Files in folder Users Files 1 week will be deleted each week. ¥ The User Interface - A quick tour. Mouse: Left Button - All commands except manual FOCUS. Right Button - Manual FOCUS only. Right Button + Shift - Manual Stigmators. Monitor: Menu Bar - pull down menus Control Areas: Settings, Imaging, Image Manipulation, Survey, (Analytical), Adjustments, Image only. Button Bar: Screen Functions, Auto-Functions, Control Areas. Command Button: Pump, Vent, HT on, etc. (turn green when on) Option Button: Scan times, Filter frames. Adjusters: Magnification, contrast, brightness (lock-in sliders) Dialog Box: Bias control for SE, SetGun dialog, etc. (has OK button and can contain check buttons, radio buttons and list box) Text Box: User can type in information. List box: Filename list, i.e. IMG, TIF.
