The Field Museum seeks candidates for a two-year post-doctoral fellowship
emphasizing interdisciplinary research as well as the central role of museum
collections. The Boyd Fellowship program was created in honor of former Field
Museum president Willard Boyd, who stressed the importance of interdisciplinary
work to the intellectual strength of the Museum. The successful candidate will
have a successful record of research in an area that bridges any of the
Museum's scientific areas, including anthropology, archaeology, botany,
systematics, evolutionary biology, conservation biology, paleontology, and
zoology. Research must involve development and use of the Museum's
collections, which span anthropology, botany, geology, zoology, as well as
tissue collections for molecular work. Boyd fellows are expected to
participate fully as part of the Field Museum academic community, to undertake
high-quality original research, and to present their work to both academic and
public audiences. The fellowship runs for two years, carries full Museum
benefits, and a modest research budget.
The Field Museum is an equal opportunity employer and actively encourages
applicants from diverse backgrounds. Send cover letter, including outline of a
project, C.V., and names of references to Chair, Boyd Post-Doctoral Fellow
Search Committee, Office of Academic Affairs, The Field Museum, 1400 S. Lake
Shore Drive, Chicago, IL 60605. Applications will be accepted until Sept. 30,
2002. Position available starting January 2003 and will remain open until
filled.

Summary of Ph.D. Thesis
Title: Profile of GTH and GH in Response to Different
Hormones, Temperature and Pesticide in the Indian
Catfish, Clarias batrachus (Linn.) During its
Reproductively Active Phases.
Hormones from hypothalamus, pituitary and gonad
regulate reproduction. Of the pituitary hormones
gonadotropins play an important role. However, in fishes
like catfish, eel etc only one gonadotropin is reported so
far. Therefore, to determine the number of gonadotropins
and the role of these gonadotropins in the reproduction of
the Indian catfish, Clarias batrachus, the present study
was undertaken. Growth hormone has also been implicated in
fish reproduction. Further, GH can be obtained from the
same lot of pituitaries used for GTH purification.
Therefore, in the present study GTH and GH were purified by
column chromatography. The purified hormones were tested
for their biological activity under in vivo and in vitro
conditions. The parameters considered were steroidogenesis,
induction of germinal vesicle breakdown (GVBD) and T3
production. Physical characterization of these fractions
was achieved by SDS-PAGE. Employing mammalian and piscine
antisera against various pituitary hormones, immunological
characterization of the purified compounds were determined.
Chemical characterization of these fractions was performed
by partial N-terminal aminoacid sequencing. Antisera of
these purified fractions were raised in New Zealand
Rabbits. Cross reactivity of the antisera was determined by
double immunodiffusion assay. Separate competitive ELISA
systems were developed for each hormone. Sensitivity of the
GTH II was 15.6pg/100ul, while that of the GH was
31.5pg/100ul. Each ELISA system was also validated by inter-
assay and intra assay variations and recovery.
Plasma levels of the GTH II and GH exhibited
significant diel variations during the reproductively
active phases. Such discontinuous secretion of these
hormones may be indicative of the variations in the
sensitivity of their receptors.
Effects of various hormones on GTH II and GH were
studied in stage specific fish. Actions of GnRH, sex
steroids, indolamines and Morphine were season dependent.
In addition to that sex steroids were found to be
modulating the actions of sGnRH. GnRH induced GH secretion
needs special mention.
Effects of pesticides belonging to organophosphate,
organochloride and carbamate were also monitored. All these
pesticide groups were found to be inhibitory to both the
GTH II and GH. However impact of carbaryl was more
pronounced than the other two.

Geology principle, Geological history, Geophysics,
Geochemistry (including environment, deposit, isotope,
petrology etc.), etc. My research interests and activities
are concentrated on igneous geological processes. During my
Master course, I have carried out a comprehensive study on
the relation between the Cenozoic volcanic rocks and the
uplift of Qinghai-Tibet Plateau in China. And firstly
discovered that there exist Adakite rocks in northern
Tibet, which is very important for the uplifted mechanism
of Qinghan-Tibet plateau. At the same time, have
investigated the ophiolite belt between Luogang-Rencuo, and
believed it is the result of expansion setting of little
oceanic basin at the beginning of after early Proterozoic.
After the completion of Masters course, I qualified the
doctoral fellowship of Institute of Geochemistry, Chinese
Academy of Sciences (IGCAS). And focussed my research
activities on the relation of medium-mafic dikes and
lithospheric extension in eastern China during Mesozoic. I
have carried out detailed and comprehensive field work in
the medium-mafic dikes of Shandong province, China. By
research, I considered that the dikes are the result of
partial melting of Enriched Mantle due to the interaction
between the subducted ancient crust (Yangtze Craton) and
the mantle peridotite. Additionally, the petrogenesis is
associated with the subduction of Ancient Ocean (Su-Lu
ocean instead of ancient pacific ocean) into NCC because of
their geochemical feature of Continental margin arc. Based
on the isotopic ages of the dikes, it is justifiable to
conclude that there exist five stages lithoshperic
extension at the least in Shandong terrain during Mesozoic.

Research Project Completed (Ph.D)
In vitro culture and Agrobacterium mediated genetic
transformation in Peanut with rice chitinase (Arachis
hypogaea L.)UK-Inidia Project for crop improvement
In vitro and genetic transformation studies in peanut
(Arachis hypogaea L.)
Arachis hypogea L. is one of the important oil seed crops
in India. It contains high protein, carbohydrate, vitamins
and minerals. The effects of amino acids (glutamine,
serine, proline, glycine and valine and additives (PVP,
Sodium citrate, activated charcoal) on plant regeneration
from cotyledon and embryonal axis explants of peanut were
studied. Cotyledon showed better response than embryonal
axis explants. The amino acids (nitrogen source) enhanced
the multiple shoot induction and additives (PVP) reduced
the phenolic substances because the legumes are
recalcitrant. Repetitive somatic embryogenesis was
achieved in embryonal axis explant with the addition of
haemoglobin. Agrobacterium transformation is influenced
by a number of factors including bacterial strain,
temperature, cell wounding, phenols, virulence genes, and
host cell division. The sensitivity of the explants to
kanamycin (selection marker) was first established before
initiating the transformation experiment in order to
determine the maximum concentration of kanamycin that
permits the regeneration of transformed shoots, while
preventing the growth of non-transformed shoots. As with
recalcitrant legumes Agrobacterium mediated transformation
of peanut appears to be cultivar and strain specific. 2.5%
of cotyledons were cocultivated and kanamycin-resistant
shoots were selected, of which 10 proved to be gus
positive. Some primary transformants were chimeric, but
others appeared uniformly transformed and showed 6.5%
segregation of gus activity in progeny. In this study,
Agrobacterium incorporated rice chitinase gene, were
transferred to peanut explants and expression was observed
in transformants.
The efficient protocols have been standardized for
organogenesis, repetitive somatic embryogenesis, factors
influencing Agrobacterium mediated genetic transformation
and chitinase gene transformation for disease resistant.
In this present investigation, chitinase gene was
introduced and expressed in the transgenic plants. These
chitinase enzymes (PR protein) confer resistance to fungal
disease called “tikka” disease in groundnut.
Arachidic acid were isolated and quandified through HPLC
and NMR and this compound were more in vitro explants.
