Poly-L-Lysine Coated Coverslips.

1. Clean coverslips with 95% ethanol and wipe with clean cheesecloth or lint free cloth.

2. Place coverslips flat in petri dish.

3. Place a large drop of 0.1% poly-L-lysine on each coverslip; withdraw fluid until a thin layer remains and let sit for 30minutes. Make sure peri dish is closed to keep a moist atmosphere. If any of the drops round up, discard that coverslip.

4. Rinse coverslip with dH20 and prop along a filter paper lined petri dish to dry. Air dry for 2 hours before introducing cells or media.

5. Store coverlsips, coated side up in a closed dish and use fresh or within 24hours.

For poly-L-lysine coated cover slips, use HMW (>300,000) poly-L-lysine Sigma Chemical Co.), at a concentration of 0.1%. Store poly-L-lsine powder at -0ºC.
0.1% Pol-L-lysine in dH20: make up small aliquots and freeze at -20ºC. Use a fresh aliquot for each coating session.
Cover slips should be coated with fresh solutions only and should be used within the same day they are coated.
